INTRODUCTION The movie Lorenzo’s oil portrays an inherited genetic disorder which is Adrenoleukodystrophy (ALD). It is a syndrome called Addison disease with an X-linked recessive gene and cerebral sclerosis (Bruckeim, A.H.M.D, the family Doctor, Educators Advantage, 1994). This disease is caused by the abnormal build-up of very long-chain of saturated fatty acids (VLCFA), hexacosanoic acid (C26:0) and teracosanoic acid (C24:0) (Moser et al, 2007) in the tissues, body fluids (Igarashi et al, 1976), the adrenal glands and brain (cerebral white matter) ,(Woodrow Wilson Biology institute,1994). On a protein level, ALD is triggered by ATP-binding cassette transporter subfamily D member 1(ABCD1) gene coding for a peroxomal membrane protein (Moser et al, 2005). Moreover, it is also caused by a single mutation at the locus Xq28 causing deficient activity of necessary enzyme ligase. The disease symptoms usually develop as a result to the abnormality of perixomes in the liver, which ultimately disrupts fatty acid metabolism and affect very long chain fatty acids causing them to be abnormal (“Lorenzo’s oil”; MCA Universal Studio,1992). Different forms of the disease Adrenoleukodystrophy can be found in three types of ALD; Adolescent cerebral X-ALD which is effective at the age of 10-21 years and cause behavioural and neurological deficits, Adrenomyeloneuropathy is effective at the age of 19-37 years and causes spinal cord paralysis and Adult cerebral X-ALD which is effective after 21…
