Introduction The purpose of this study is to study and identify four unknown organisms that fall under the following categories: Fungi, Protista, Bacteria, and Fresh Water Green Algae. These organisms fit in one of the three domains: Archaea, Bacteria, and Eukarya. Archaea and Bacteria fall under the category Prokaryotic. Of the organisms studied Bacteria falls under the Prokaryotes in the Bacteria domain. Bacteria tend to be unicellular, they have a cell wall but lack a nucleus as well as other membrane organelles, and they contain a nucleoid where all the chromosomes are located. Their mode of locomotion is thru

If these organism are what they are suspected of being then the bacteria size of the suspected bacteria should fall under the size range 0.5 micrometers to 15 micrometers? Based on bacteria’s characteristic cilia or flagella should be visible under microscopic view. Green Algae under microscopic view size would range at least 10 micrometers, the suspected Protista cells size should range between 0.8 micrometers to 2 millimeters, and most Protista cells have a light detector if the cell is a Protista then it should be attracted to the light of the microscope when viewed under the microscope. Green Algae as its names states should appear green under the microscope and its size should range between 10 micrometers to 30 centimeters. Out of all the organisms studied Green Algae should be the biggest of all. If the organisms suspected of being Yeast is in effect yeast than its cell sizes should vary between 2 micrometers to 10 micrometers. Size is a huge indicator of the cells identity as well as the color of the organisms, based on these two factors these

During the phase and process of the bacterial unknown project, vast amounts of test were performed in order to determine an unknown species of bacteria. Within the many procedures of identifying the unknown bacteria, I was first given a tube with the unknown bacteria, from which later on I started subcultures by streaking bacteria from the original tube onto a tryptic soy agar (TSA) slant. Throughout the process of identifying the unknown, new subcultures were produced in order to use the newest bacteria to conduct the tests. Thus, this process allowed the unknown bacteria to grow and maintain. Before a series of test were conducted, first a gram stain test was performed in order to determine the unknown’s gram reaction and its cell morphology.…

The focus of this lab was to identify an unknown organism based on its characteristics and the results from each of the tests. There will be various of test to choose from in order to identify the unknown organism, which will eliminate numerous possibilities and narrow it down to one. All the fundamental skills that we have learned and practiced in the lab will be used to perform on our unknown such as aseptic technique, microscopic examination, the use of differential media, and determining if it’s positive or negative. Performing aseptic techniques is the most crucial step that requires the utilizing of transferring, inoculating, and storing bacterial cultures and media. Aseptic technique is defined as procedures that prevent contamination…

To determine the unknown’s morphology is the first step in narrowing down possible organisms. A MacConkey and Mannitol Salt Agar tests were conducted on the organism and revealed the following. No growth on the MacConkey plate resulting in a gram positive bacteria and the Mannitol Salt Agar result showed it could be Staphylococcus or Micrococcus. (1) A gram stain test was used in determining the shape, size and arrangement of the unknown bacteria (1). The unknown organism is heat fixed onto the slide, in order for the organism to be stained.…

To start the process of identifying the unknown bacterium following Bergey’s Manual, a broth, a deep, an agar slant and a streak plate are all inoculated with the unknown bacterium via a needle for the deep and a loop for the broth, agar slant and streak plate. After forty-eight hours all the mediums are examined to see what growth has occurred and the characteristics of growth (if any) are documented. This bacterium was found to be motile from results via the deep, a thin film was visible atop the broth, and sediment was seen at the bottom, with some growth on the agar slant. The growth on the slant appeared to be white in color; almost clear and spiny in its pattern of growth. The growth of bacteria on the streak plate was also successful.…

The study of microbiology requires not only understanding the microscopic organisms, but also the understanding of lab techniques and procedures used to identify, control, and manipulate microorganisms. The identification of microorganisms is not only important in microbiology lab, but also in the medical field to identify an agent of a disease that will help treat the patient by using the correct antibiotics to kill off the host. In this unknown lab report, techniques and procedures learned in the microbiology laboratory during the semester that was performed to test ones practical understanding of microbiology. The sole purpose of the unknown lab is to demonstrate understanding of the experimental methods and lab techniques learned during…

Biochemical Unknown I. Introduction Cultural Characteristics or morphology and biochemical tests can be used to identify and classify microorganism. By culturing microorganism in nutrient broth, slants, and on nutrient agar plates, the cultural characteristics or morphology can be determined. There were multiple tiny whittish isolated colonies present, all of which showed the same growth pattern. Three colonies were chosen at random and Gram stains were performed on each (procedure for the Gram stain was used according to laboratory manual ex. 9). Each of the three Gram stains showed positive rods. After the Gram stain was determined, I began to do more research on microorganisms that had that identification and narrowing down all the possible tests to continue for identifying. Specific biochemical tests were performed.…

Using the Bergey’s Manual to identify my unknown bacterium. The Bergey’s Manual is the collaborative effort of the numerous individuals (Alverno College, 2014). First thing that will help me to identify my unknown bacteria is if its gram-negative or gram-positive; also, looking at the shape to help with the identification, so shapes such as rods or cocci. Referring back to the gram-staining on my results, it was concluded that my unknown is gram-negative since during the safranin wash, the bacterium turned purple to red (Lee, 2017). The shape of my unknown bacterium according to the microscope observation at 100x, is seen to be a coccus shape.…

Identification of unknown organisms is useful in a wide variety of settings; from clinical diagnostics to environmental monitoring (1). Species of bacteria can differ fundamentally in aspects ranging from morphology to metabolism. Exploiting these differences can allow for identification of unknown organisms. Gross morphological characterizations are crude, and better replaced by more accurate and specific tests. At a fundamental level, tests such as the Gram stain divide bacteria in to two broad categories based on the characteristics of their cell wall (2).…
