Where is color in basic stain? |
in positive ion |
Where is color in acidic stains? |
in negative ions |
What charge does bacteria have? |
negative |
What type of stain would stain the bacteria itself? Why? |
basic stain opposites attract; positive ions would be attracted to negative bacteria |
What type of stain stains the background instead of the bacteria? |
Acidic stains |
What are the 5 Basic stains? |
1. methylene blue 2. crystal violet 3. malachite green 4. safranin (red) 5. carbol fuchsin (red) |
What are the 2 acidic stains? What color are they? |
- nigrosene - india ink dark blue to black |
Capsule stain |
uses both a basic and an acidic stain but capsule is electrically neutral so it won't stain at all, will remain white/colorless |
How did we prepare simple stain |
1. 1 loopful of water with 1 loopful of B. megaterium. smear it around 2. let air dry 3. heat fix 4. stain with METHYLENE BLUE (basic stain) for 1 min 5. rinse and blot 6. observe |
How did we prepare negative stain? |
1. 1 loopful of nigrosine (acidic stain) and B. mega at END of SLIDE 2. slide spreader 3. air dry 4. observe |
How did we prepare capsule stain? |
1. 1 loopful India ink (acidic stain) with B. mega 2. slide spreader 3. air dry 4. heat fix 5. counterstain with Crystal violet (basic stain) for 1 min 6. rinse and blot 7. observe |
two reasons we heat fix slides? |
1. kill organisms on slide 2. make organisms adhere to slide |
Why don't we need to heat fix the negative slide? |
It doesn't get rinsed |
What did the basic stain of Bacillus megaterium (underlined) look like? |
blue streptobacillus on white/clear background |
What did the negative stain of Bacillus megaterium (underlined) look like |
White/clear streptobacillus with dark/black background? |
What did capusle stain of Bacillus megaterium (underlined) look like? |
Black/dark background Purple bacteria surrounded by 'white clouds' which are the capsules |
Compare negative stain vs. capsule stain of B. mega |
in negative stain, the whole bacillus was almost glow in the dark white. With capsule staining, only the 'edges' of the bacillus are glow in the dark white. |
