The proton pump inhibitors are absorbed in the small intestine.
They are brought to the parietal cells via the systemic circulation. They are activated in the acid environment of the gastric parietal cell secretory canaliculi. The active metabolites bind covalently to the H+-K+-ATPase thereby irreversibly inhibiting acid release. note: only the charged drug does this |

PPIs are usually taken 0.5-1 hour before meals.

Eating stimulates pump activity and acid production
Acid is required for activation of the drug. The pumps need to be active at the wall of the canalicular space to bind PPIs. |
