Homogenate Lab Report

Improved Essays
Homogenates of liver were provided. The homogenates were made by homogenizing liver in a 1:20 w/v (1g: 20 ml) ratio in a sucrose-phosphate buffer, in which 250mM of sucrose and 50mM NaPO, with a pH of 7.4 were added. This information was used to conduct an experiment to determine the concentration of DNA, protein, and carbohydrates in different homogenate samples, including heart, liver, and kidney of Bos taurus, while also looking for the diverse protein sizes found in each homogenate sample. The liver homogenate was then stored at -70°C. Of 300 µl, 5 µl of homogenate was placed into 2 µl of protein gel sample buffer. The homogenate was then mixed and heated at 80°C for 10 minutes. After heating, the sample was placed in a microcentrifuge

Purpose and Background Cells divide in two ways, mitosis and meiosis. Mitosis is used to produce cells that are genetically identical to the parent cell for growth, asexual reproduction, or repair after injury. Cells that are produced by mitosis are diploid, meaning that they have two complete sets of chromosomes, one from each parent. Meiosis is used to produce haploid cells that have only one set of chromosomes, a mix of chromosomes from both parents. Meiosis produces cells that are genetically unique from their parent cells.…

Lab #5: Introduction to Metasploit on Kali Linux Team: CRYPTERS 1 d. Why is it usually a bad idea to operate in the Linux environment as root? If you are unfamiliar with the concept of the root user, do a quick google search. It is always a good practice on any operating system to run your applications on a user level and leave the administrative tasks to the root user, and only on a per-need basis. Applications are meant to be run by users with non-administrative privileges.(Power December 4, 2010)…

Protein Purification III Parts 09/15/2015, 09/22/2015, 09/29/2015 Kaisha Butz Lab Partner: Ryan Van Abstract The lab was comprised of three parts, the purpose of which was to separate a protein mix and analyze the results. The first part was separating the proteins; the second part was reacting the proteins with an enzyme, and the third part was analyzing each fraction from Part I to see if separation was successful. It was hypothesized that out of the four proteins in the mix that the glucose oxidase which had the greatest negative charge out of the four proteins would elute last.…

Salcha-1 was incubated in six Erlenmeyer flasks (1L) containing 200mL of Potato Dextrose Broth (10g/ L Great Value mashed potatoes with 5g/L D-glucose). One of the five flasks received approximately 1.0-1.3g of the following metal compounds: cobalt (II) chloride hexahydrate, cobalt (II) nitrate hexahydrate, cobalt (II) thiocyanate, erbium chloride hexahydrate, or palladium (II) acetate. The sixth flask was reserved as a control and only contained PDB. Salcha-1 was incubated on an Innova 5000 shaker at 135rpm for three weeks at room temperature.…

Studying the reaction of the enzyme catalase in different temperatures The purpose of this experiment was to measure the speed and the effectiveness of the enzyme catalase. The enzyme catalase purpose is to break down harmful substances and turn them into oxygen and water. The substance in this environment was hydrogen peroxide, and the study was to see how well the chicken liver would work if it was in a boiled environment, room temperature environment, an acid immersed environment and a basic environment.…

The purpose of this lab was to use NMR, Recrystallization, TLC, and Melting point to separate and identify the components of a given unknown mixture. The first step of the experiment was to use the two NMR spectra given to identify the major and minor components of the unknown mixture. When an external magnetic field is applied, there is a quantized interaction between the magnetic field itself and the magnetic moment of the proton. Thus, the magnetic moment of the proton either applies with or against the magnetic field depending on direction the proton is spinning.…

The purpose of this experiment was to determine the concentration of succinate dehydrogenase in six fractions made by students. Succinate dehydrogenase is an enzyme located in the mitochondrial membrane and aids in cell respirations and enzyme generation. In order to conduct this experiment, students cut a liver and through a process created six fractions. Each of the six fractions was moved to a cuvette with solutions containing different volumes and recoding the absorbance every five seconds for four minutes. Once the absorbance was obtained for each, we determined that cuvette E (0.0012) had the least activity per milliliter while cuvette B had the most activity per milliliter 0.0138 (Table 2).…

The aim of this experiment was to determine the enzymatic activity of succinate dehydrogenase (SDH) in various solutions, specifically those containing and not containing a reagent. The assigned reagent was malonate, which was previously hypothesized to be an inhibitor of SDH. Bovine liver was manipulated primarily through centrifugation, and mitochondrial fractions were then created to serve as the enzyme-rich solutions the reagent would act upon. To determine the protein concentration of the mitochondrial fraction, the Bradford assay was implemented; the result will later be used in the calculation of the specific activity of SDH.…
